“Biology of Vascular Smooth Muscle: Vasoconstriction and Dilatation”
The discovery that the vascular contractility is regulated by a potent diffusible factor derived from the endothelium made by Furchgott and Zawadzki in 1980 is an epoch-making milestone in vascular biology. For a long time, the endothelium has been largely considered as a selective permeable barrier between the circulating blood and the vascular smooth muscle. Intensive studies evoked by Furchgott and Zawadzki’s work since then have firmly established that the endothelium-derived nitric oxide, prostanoids, endothelin, and endothelium-dependent hyperpolarization play a dominant role in the regulation of vascular activities both under physiolog- ical and pathophysiological conditions.
Meanwhile, aided by the rapid advances in biology and medical science in particular advance in molecular biology, numerous progresses have been made on the mechanisms underlying the vascular motion, including STIM–Orai protein-mediated store-operated Ca2+ entry, RhoA-Rho kinase signaling, the regulation of myosin light chain phosphatase activity, the effect of mitochondria-derived reactive oxygen species, and redox modulation of protein activities.
